CJC-1295 with DAC vs without DAC: what is the difference?

In brief

Both names refer to synthetic analogues of the GHRH(1-29) fragment carrying four amino-acid substitutions. The difference is the DAC (Drug Affinity Complex) component: a linker that binds the peptide to blood albumin and extends its action from minutes to several days. "CJC-1295 without DAC" is the same modified GRF(1-29) without the linker — a short-acting analogue. The names are constantly confused, but these are different molecules with different pharmacokinetics.

The D-amino acid means mass does not confirm stereochemistry.

The shared backbone

Both compounds are built on fragment 1-29 of growth-hormone-releasing hormone (GHRH) — the shortest segment retaining the full hormone's biological activity. In both versions it carries four amino-acid substitutions that increase resistance to enzymatic degradation, hence the literature name "tetrasubstituted GRF(1-29)".

What DAC does

DAC (Drug Affinity Complex) is a maleimidopropionic-acid-based linker that covalently couples the peptide to albumin in blood. Albumin binding protects the peptide from rapid clearance: in the clinical study by Teichman and colleagues (2006), single administrations of CJC-1295 with DAC sustained elevated growth-hormone and IGF-1 levels in healthy adults for several days.

The no-DAC version lacks the linker and is short-acting — on the scale of minutes, like GHRH itself. Substantially fewer clinical data exist for this version specifically.

Why the names are confused

Historically, "CJC-1295" was the development code for the DAC version specifically. The research-materials market later adopted "CJC-1295 without DAC" and "Modified GRF (1-29)" as interchangeable labels for the short version — usage documented in a public FDA docket. When reading literature or documentation, always check which of the two molecules is meant: their masses and pharmacokinetics differ.

Catalogue position

The Moscow Peptides catalogue item is the no-DAC version (Modified GRF 1-29). Batch identity can be confirmed only through reviewed analytical documentation; until then, identity remains pending confirmation. The material is for laboratory research only.
